Its a word from HipHop. It define the last line of a rhyme, the one that bring it on point. If u bring good punch lines the crowd will scream.
Punch Lining
The act of repeating the funniest part of a joke or movie quote to your friends after they have all laughed at it, causing more laughter among the group.
Example:
Jeff: "dude that joke Chad told was hilarious."
Greg: "oh yea we were punch lining that one all night, it never gets old."

Prepared rhymes/Bars thatchu kick in a battle. You could say the opposite of a freestyle. sometimes it's hard to tell wether it's freestyle or if its a punch line.
Example:

After a few minutes the young man burst into the room where the boss was and exclaimed, "Boss! That woman has a shrimp stuck in her vagina!". The boss was shocked "No way! I've gotta see this.". So they both went to see the body and the young man pointed "You see...". The boss chuckled to himself. "That's no shrimp. That's a clitoris". The young man, was confused. "A clitoris? Well it sure tasted like shrimp!"

Punch Line Killer
Someone who says something stupid at the end of a good joke and ruins it for everyone. Replaces the punch line leaving the joke useless.

When a person not involved in telling joke interrupts the person who is telling joke, thereby flummoxing the person and causing the mistiming of the punch line.
